Auxin has a crucial influence on genes expression. In this work, we show that transcriptional activity of an auxin-sensitive gene MAKR4 (MEMBRANE ASSOSIATED KINASE REGULATOR 4) depends on the composite elements in its upstream regulatory region. Analysis of such elements could clarify the understanding of the regulation of this gene and other auxin-sensitive genes.
Thank you for the presentation, I have 2 questions regarding slide 5.
1) Are the plants in third and the fourth pictures heterozygous for the mutations? If so, do they correspond to any of the boxes in slide 6?
2) In your confocal images, I can see that there is no auxin induction in your mutant lines. Nevertheless, the background level of GFP expression (in mock samples) differs from line to line. Is it due to different microscope settings or due to the genome location of construct insertions? Can it affect your results?

1) These pictures illustrate the pattern we observe in lines with mutated sites (one mutated site or two and three sites at once). All lines we examined are homozygous for the mutations, except for the line with mutated second site.
2) Basic GFP level might depend on the location of construct insertions, but it doesn’t affect results, because then we compare different lines, we normalize each one of them on the background GFP expression level.

1) Do other MAKRs (members of MEMBRANE ASSOSIATED KINASE REGULATOR family) are auxin-sensitive genes?
2) Why did you choose MARK4 for your analysis? Is it more important or interesting then other MAKRs?

1) Not all MAKRs are auxin-sensitive, but there are two auxin-sensitive members of this family – MAKR4 and MAKR6, both of them are expressed in the Arabidopsis root;
2) MAKR4 is the marker of lateral root formation in Arabidopsis; we were interested how auxin mediates expression of this gene.
